Protein extraction, a key to success in Western blot

The diversity of tissues and organisms which are analyzed by plant science researchers is quite impressive. From leaf, root, flowers, fruits and seeds, to algal and diatom cultures. Oftentimes the same extraction protocol cannot be applied to all samples which are to be analyzed in the lab, and efficient protein extraction is the first step to a successful detection of a target protein in Western blot.
 
The basics for proetin extraction are: use of suitable buffers with protease inhibitors and, in some cases, proteasome inihibitors. All work has to be done very promptly and effectively, and the best results are obtained from freshly extracted samples. Even light conditions during extraction may influence the outcome, especially for proteins which degrade in the light, like ELF3. In such a case, a darkroom is a must.
